The aging of the skin is an inevitable physiological process that results in the loss of its natural elasticity and the appearance of wrinkles. H2DCF-DA exhibits high sensitivity to hydrogen peroxide (H2O2) and can be applied to detect intracellular production of reactive oxygen species (ROS).
By measuring the intensity of DCF fluorescence, we can assess the level of ROS generation within the cells.
During the cultivation period, samples from the PROTAI™ series were added to the culture medium (10 μl) and cultured for 1 hour under light-avoiding conditions at 37°C.
After cultivation, cells were washed with PBS, and then the fluorescence signal was determined using a fluorescence spectrophotometer (excitation wavelength/emission wavelength: 485/538 nm). All data are the averages of three repeated experiments.
After incorporating PROTAI™, the reduction in ROS (Reactive Oxygen Species) free radicals is 15%.

Through experimental validation, it has been shown to effectively promote the migration of epithelial cells, thereby facilitating wound repair and enhancing skin moisture and elasticity.
With the addition of PROTAI™, there was a significant increase in migration repair,
ranging from 77 - 100 %, demonstrating pronounced healing effects.
The single-molecule membrane of HaCaT epithelial cells was employed for the detection of wounds caused by incisions. Pre-treatment images were obtained before the application of PROTAI™. After 18 hours of cell incubation, images were captured following fixation and post-processing.
By comparing and analyzing the images before and after through microscopic imaging, the central gap area was identified.
After 18 hours of migration repair testing, samples treated with the sequence-specific peptide of PROTAI™
showed a reduction of 3.4 ± 1.1.1 in the blank area, representing a decrease of 77%.
Following treatment with a high concentration of PROTAI™, the blank area decreased by 0.7 ± 0.33, reflecting a reduction of 95%.
In summary, based on these results, PROTAI™ demonstrates outstanding effects in inducing epithelial cell migration and promoting skin repair.
